Behavior of ERK-PP under various inhibitory conditions. When the phosphorylation reaction of EGFR alone is targeted by kinase inhibitors (Single inhibition), when phosphorylation of EGFR and association of GAP are targeted (Multiple inhibitions: upstream targets A), when phosphorylation of EGFR and several other association reactions in the Grb2 module are targeted (Multiple inhibitions: upstream targets B), and when association reactions in the Ras Shc-dependent, Raf, MEK, and ERK modules are targeted (Multiple inhibition: downstream targets). Various levels of inhibition are represented by the green (10%), blue (30%), red (50%), yellow (70%) and brown (90%) lines.
